Innate Immune DNA-Sensing Pathways

ثبت نشده
چکیده

Recognition of foreign DNA in intracellular compartments or in the cytoplasm sounds an alarm bell signaling pathogenic invasion. In response, innate immune DNA-sensing pathways launch an antimicrobial type I interferon (IFN) response, which becomes damaging to the host if unregulated or inappropriately activated by self-DNA. This Select highlights recent insights into activation and regulation of innate immune DNA-sensing pathways.

Stimulation of cytoplasmic DNA sensing pathways in vitro and in vivo.

In order to efficiently stimulate an innate immune response, DNA must be of sufficient length and purity. We present a method where double stranded DNA (dsDNA) which has the requisite characteristics to stimulate the cytoplasmic DNA sensing pathways can be generated cheaply and with ease. By the concatemerization of short, synthetic oligonucleotides (which lack CpG motifs), dsDNA can be generat...

Targeting Cytosolic Nucleic Acid-Sensing Pathways for Cancer Immunotherapies

The innate immune system provides the first line of defense against pathogen infection though also influences pathways involved in cancer immunosurveillance. The innate immune system relies on a limited set of germ line-encoded sensors termed pattern recognition receptors (PRRs), signaling proteins and immune response factors. Cytosolic receptors mediate recognition of danger damage-associated ...

Evasion of innate cytosolic DNA sensing by a gammaherpesvirus facilitates establishment of latent infection.

Herpesviruses are DNA viruses harboring the capacity to establish lifelong latent-recurrent infections. There is limited knowledge about viruses targeting the innate DNA-sensing pathway, as well as how the innate system impacts on the latent reservoir of herpesvirus infections. In this article, we report that murine gammaherpesvirus 68 (MHV68), in contrast to α- and β-herpesviruses, induces ver...

The role of innate immune signals in antitumor immunity

Innate immunity serves as a first line of defense against infectious agents, and germ-line-encoded pattern recognition receptors detect stressed and infected cells and elicit potent effector activities that accomplish efficient microbe containment. Recent evidence demonstrates that these pattern-sensing systems are also applicable to the recognition of tumor-derived stress-related factors. In p...

RIGulation of STING expression: at the crossroads of viral RNA and DNA sensing pathways

The innate immune sensing of pathogens is important for host to mount defensive responses. STING has emerged in recent years as a critical signaling adaptor in the immune response to cytosolic DNA and RNA derived from pathogens. Liu et al. (2016) demonstrate that the RIG-I-dependent RNA sensing signaling induces STING expression via a TNF-α and IFN-α synergy. The up-regulation of STING is vital...
